Description: Raffinerien werden in naher Zukunft vor der Aufgabe stehen, in größerem Umfang Rohöle minderer Qualität und Erdöl-Vakuumdestillationsrückstände (Gudron) in hochwertige mittlere Destillate umzuwandeln. Berichtet wird über den aussichtsreichen Einsatz von öl- oder wasserlöslichen Precursoren für die Erzeugung hochdisperser Metallsulfid-Katalysatoren im Hydrocrackprozeß von Gudron. Tabelliert sind chemische Zusammensetzung und Kennzahlen vom eingesetzten Gudron. Zunächst erfolgten Batch-Autoklavenversuche mit öllöslichen W-, Mo-, Co-, Ni-Naphtenaten als Precursoren. Die Ergebnisse sind tabelliert. Röntgendiffraktometrische Rückstandsuntersuchungen ergaben, daß NiS, CoS, MoS2 und WS2 in Form von Kristalliten gebildet worden waren. Es folgten Tests mit einer kontinuierlichen Versuchsanlage in einer Reaktorkaskade (Schema). Bei den Batchversuchen erwiesen sich besonders wirksam die Systeme NiW und CoMo, die bei hohen Umsätzen und geringen Koksbildungen auch eine hohe Entschwefelung ermöglichten. Der Einfluß wichtiger Prozeßparameter auf das Hydrocracken wurde in Batchversuchen am Beispiel von NiW-Sulfid-Katalysatoren untersucht; die Ergebnisse sind in sechs Diagrammen dargestellt. Es zeigte sich z.B., daß der Katalysator den Umsatz nicht nur fördert, sondern ihn sogar verringert. Jedoch findet gleichzeitig eine signifikante Abnahme der Koksbildung statt. Bimetallische Sulfidsysteme wie NiS/WS2 oder CoS/MoS2 haben sich besonders leistungsfähig erwiesen. Durch den kontinuierlichen zweistufigen Prozeß wird die Koksbildung gesenkt und mehr leichtes und mittleres Destillat erhalten. Dabei wird zunächst bei 〈 420 Grad C ein teilweiser Umsatz erreicht und in der zweiten Stufe bei 440 Grad C bis 455 Grad C der integrale Gudron-Umsatz auf fasr 90 % erhöht, ohne drastischen Anstieg der Koksbildung.

Description: Background: FoxO3a is a transcription factor crucial in regulation of cell metabolism, stress resistance and immunity. Recently polymorphisms of FOXO3a have been associated with favorable outcome in inflammatory disease. However, mechanistic insight in FoxO3a effects is still limited. Here, we investigated the role of FoxO3a on NK-cell responses in viral myocarditis.Methods: A mouse model of Coxsackie Virus B3 (CVB3) myocarditis in wild-type and FoxO3a-/- mice was used to investigate the effects of FoxO3a on viral load and inflammation. Characterization of NK cells was performed by cytotoxic assay, surface marker- and interferon expression.Results: FoxO3a-/- mice showed significantly reduced myocardial inflammation, lower viral titers and attenuated expression of pro-inflammatory cytokines compared to wild-type mice 7 days post infection. Viral titers were significantly lower in FoxO3a-/- mice at day 3 while IFN γ and NKp46 expression were up-regulated within the heart suggesting early viral control by enhanced NK cell activity. There were no differences in expression of CAR (coxsackie-adenovirus receptor). Additionally NKp46+ splenic NK cells of FoxO3a-/- mice exhibited a superior functional status ex vivo determined by enhanced expression of CD69, higher frequencies of CD11b+CD27+ effector NK cells and increased cytotoxicity compared to NK cells of wild-type littermates. These effects were not due to different levels of apoptosis. MicroRNA-155 expression, recently reported to be essential in NK-cell activation, was significantly elevated in FoxO3a-/- NK cells while its inhibition led to diminished production of IFNγ in Foxo3a deficient animals. Moreover analysis of healthy human donors for the gain of function FoxO3a SNP rs9400239 revealed significantly reduced CD107a dependent degranulation activity in donors homozygous for the allelic variant.Conclusion: Our results implicate FoxO3a directly in regulation of NK-cell function and suggest an important role in innate immune responses to viral infection. While the anti-inflammatory effects of FoxO3a seem beneficial in chronic inflammation the suppression of innate immunity in acute viral myocarditis could have detrimental effects.

Description: Coxsackievirus B3 (CVB3) is a major heart pathogen against which no therapy exists to date. The potential of a combination treatment consisting of a proteinaceous virus receptor trap and an RNA interference-based component to prevent CVB3-induced myocarditis was investigated. A soluble variant of the extracellular domain of the coxsackievirus-adenovirus receptor (sCAR-Fc) was expressed from an adenoviral vector and 2 short hairpin RNAs (shRdRp2.4) directed against CVB3 were delivered by an adeno-associated virus (AAV) vector. Cell culture experiments revealed additive antiviral activity of the combined application. In a CVB3-induced mouse myocarditis model, both components applied individually significantly reduced inflammation and viral load in the heart. The combination exerted an additive antiviral effect and reduced heart pathology. Hemodynamic measurement revealed that infection with CVB3 resulted in impaired heart function, as illustrated by a drastically reduced cardiac output and impaired contractility and relaxation. Treatment with either sCAR-Fc or shRdRp2.4 significantly improved these parameters. Importantly, the combination of both components led to a further significant improvement of heart function. Combination of sCAR-Fc and shRdRp2.4 exerted additive effects and was significantly more effective than either of the single treatments in inhibiting CVB3-induced myocarditis and preventing cardiac dysfunction.
